    Excess Iodide Effects on Autophagy of Human Nerve Cells (SH-SY5Y)

    • Objectives To investigate the damage of excess iodide on human neuroblastoma cells (SH-SY5Y)and its related mechanism.Methods SH-SY5Y cells were treated with potassium iodide (KI)at the concentrations of 0, 10, 20 and 30 mmol/L in the medium for 24 hours.The viability of cells, the generation of autophagic vesicles and the expression of Beclin1 were tested by CCK-8 assay, monodansylcadaverine (MDC)-staining assay and western blot assay.Results Compared with the control group, the viability of cells in the 30 mmol/L KI group was decreased obviously (P<0.05), the generation of autophagic vesicles was increased (P<0.05)and the expression of Beclin1 protein was higher significantly (P<0.05).Conclusions Excess iodide can notablely decrease the viability of SH-SY5Y cells, increase the generation of autophagic vesicles and increase the expression of Beclin1 protein in SH-SY5Y cells. These results suggested that the damage of nerve cell induced by excess iodide might be related to autophagy.
