    Relationship between Burden of BaP in Maternal and Cord Serum and PAH-DNA Adducts in Cord Blood

      目的 了解B市母婴多环芳烃负荷水平与胎儿脐带血DNA加合物之间的关系。
      方法 于2012年7月至12月收集B市某医院产前孕妇外周静脉血(n=216) 及新生儿脐带血(n=193), 并选取185对匹配的产妇与新生儿样本进行分析。利用高效液相色谱-荧光检测法测定母血和脐带血清中的苯并(a)芘(BaP)浓度, 采用毛细管电泳-激光诱导荧光检测部分脐血中的BPDE-DNA加合物的浓度。
      结果 母血和脐带血血清BaP的检出率分别是47.3%和49.7%。脐带血中血清BaP浓度高于母亲血清(P=0.002);胎儿脐带血中的BPDE-DNA加合物的检出率是86.1%, 但脐带血中的BPDE-DNA加合物与脐带血血清BaP水平无明显相关性。
      结论 该市母婴血清中均能检测到致癌性多环芳烃, 证实了多环芳烃的跨胎盘暴露已造成发育中的胎儿DNA的早期损伤。

       

      Abstract:
      Objectives To investigate the burden of BaP in pregnant women and newborns and to discuss the relationship between BaP in cord serum and BPDE-DNA adducts in cord blood.
      Methods The peripheral venous blood of 216 pregnant women during prenatal check were collected from a hospital in B city and their behavior and life style were investigated by questionnaire survey in July to December in 2012.The content of serum BaP was detected by high-performance liquid chromatography (HPLC) and the concentration of BPDE-DNA adducts in cord blood was determined by capillary electrophoresis-laser induced fluorescence immunoassay.The results in 185 paired maternal and cord samples were analyzed.
      Results The detection rate of BaP in maternal and cord serum samples is 47.3% and 49.7% respectively. BaP in cord serumwas higher than that in paired maternal samples (P=0.002), but the concentration of PAH-DNA adducts in cord blood are not statistically associated with the content of BaP either in cord or maternal serum.
      Conclusions The carcinogenic BaP can be detected in maternal and cord serum. The DNA damage in fetus through transplacental exposure to BaP is also confirmed.
