    顶空气相色谱法测定尿中三氯乙醇

    Determination of Trichloroethanol in Urine by Headspace Gas Chromatography

      目的 建立尿中三氯乙烯代谢物三氯乙醇的顶空气相色谱测定方法。
      方法 尿样经β-葡萄糖苷醛酸酶水解后, 利用顶空法进样, FFAP毛细管柱分离, 电子捕获检测器检测尿中三氯乙酸总量, 以保留时间定性, 峰面积定量。研究过程中根据正交试验结果, 选择最优的实验条件。
      结果 方法的最低检出浓度为0.03 μg /mL, 尿中三氯乙醇浓度在0.03 ~ 155 μg /mL范围内线性关系良好, 相关系数0.9996, 方法的相对标准偏差范围为4.1% ~ 10%, 回收率在87% ~ 108%之间。
      结论 该方法灵敏度较高, 精密度较好, 能够用于尿样中三氯乙醇的测定。

       

      Abstract:
      Objectives To develop an analytical method of using headspace gas chromatography for trichloroethanol, the metabolite of trichloroethylene, in urine sample.
      Methods Glucuronate-trichloroethanol in the urine of workers exposed to trichloroethylene was hydrolyzed by β-glucuronidase.Then the total trichloroethanol in sample was determined by headspace Gas Chromatography.Orthogonal analysis was used to optimize the best experimental condition and the best effective level.
      Results The minimum detectable concentration of trichloroethanol in urine was 30 μg /L.The linearity relationship of trichloroethanol in urine was in the range from 0.03 μg /mL to 155 μg /mL.Coefficient correlation of the method was 0.9996.The relative standard deviation was from 4.1% to 10% while the recovery rate was in the range from 87% to 108%.
      Conclusion This method was precise and sensitive.It could be used for the detection of trichloroethanol in urine.
