    茶皂素对大鼠亚慢性毒性作用的研究

    Subchronic Toxicity of Tea Saponin on Rats

      目的 研究茶皂素对大鼠的亚慢性毒性作用。
      方法 选88只SPF级Wistar大鼠随机分为茶皂素高(500 mg/(kg·bw))、中(100 mg/(kg·bw))、低(20 mg/(kg·bw))剂量组和对照组(无菌蒸馏水), 每组22只, 雌雄各半。每天经口灌胃染毒1次, 连续90 d。实验期间, 及时记录大鼠一般状况、动物体重及食物消耗量, 实验末期进行血常规、血生化和尿液指标检查, 解剖动物进行病理观察, 计算脏器系数。
      结果 实验期间, 各组动物均无死亡情况。低剂量组动物, 未见明显异常情况。雄鼠中剂量组的肺脏器系数明显增高(P<0.05), 雄鼠高剂量组的体重与对照组相比明显降低(P<0.05), 而血常规(白细胞)、脏器系数(脑、脾、肺)与对照组相比明显增高(P<0.05);雌鼠高剂量组的生化指标(谷丙转氨酶、碱性磷酸酶、血清胆碱酯酶)与对照组相比明显增高(P<0.05), 而白蛋白与对照组相比降低(P<0.05)。
      结论 本实验显示茶皂素(30%原药)对动物体重有一定影响, Wistar大鼠亚慢性经口毒性实验的最大无作用剂量为20 mg/(kg·bw), 观察到有害作用的最小剂量为100 mg/(kg·bw)。

       

      Abstract:
      Objectives To explore the subchronic toxicity of tea saponin on rats.
      Methods Eighty eight Wistar rats (half male and half female) were divided into 4 groups:a control group and 3 saponin groups (high, medium and low-dose group). Animals were treated once daily by gavage for 90 days. Clinical signs, body weight, food consumption, organ weight, serum biochemistry, urinalysis, and histopathology were observed and recorded.
      Results No death was observed in all rats. No abnormalities was found in the low-dose group.In comparison with the control group, the lung index of males in the medium-dose group was higher (P < 0.05);the body weight of males in the high-dose group was significantly lower (P < 0.05);the count of white blood cells, index of brain, spleen and lung in the males of high-dose group was significantly higher (P < 0.05), the ALB concentration of females in the high-dose group was lower, the activities of ALT, ALP and CHE of females in the high-dose group were significantly higher (P < 0.05).
      Conclusions The Results suggested that tea saponin might have potential impacts on the body weight of rats. In the subchronic toxicity test of tea saponin induced by oral exposure, the no observable adverse effect levels (NOAEL) was 20 mg/(kg·bw) and the lowest observed adverse effect level (LOAEL) was 100 mg/(kg·bw).
