    Effect of Prenatal Benzo (a) pyrene Exposure on Global DNA Methylation of Fetus

      目的 探讨苯并芘暴露对胎儿基因组甲基化的影响。
      方法 用高效液相色谱法检测27份新生儿脐血血清中的苯并(a)芘(BaP)浓度, 并按照BaP浓度将样本分成不同组, 采用液相色谱串联质谱(LCMS/MS)方法检测相应脐血中全基因组甲基化水平, 分析不同质量浓度BaP暴露水平与胎儿基因组甲基化水平的关系。
      结果 27份脐带血血清BaP平均质量浓度为0.185±0.1405μg/L。按照血清暴露水平分成不同水平暴露组, 随着暴露水平的增加, 相应的全基因组甲基化的水平有降低趋势。
      结论 宫内BaP暴露可能会降低胎儿基因组甲基化的水平。

       

      Abstract:
      Objectives To explore the effect of prenatal benzo (a) pyrene exposure on the global DNA methylation of fetus.
      Methods The content of BaP in cord serum of 27 newborns was detected by using high--performance liquid chromatography (HPLC)/fluorescence. The global DNA methylation were quantified by liquid chromatography--tandemmass spectrometry (LC-MS/MS). The relationship between BaP exposure level and the status of global DNA methylation in cord blood were analyzed.
      Results The average exposure level of BaP in 27 cord serum samples was 0.185 ±0.1405 μg/L. The samples were categorized into 5 groups according to BaP levels. The levels of global DNA methylation showed a negative correlation with the increase of BaP exposure.
      Conclusions Prenatal BaP exposure was associated with lower global DNA methylation.
