    大气颗粒物暴露对大鼠心肌组织氧化损伤研究

    Oxidative injury of atmospheric particulate matter on myocardium tissue of rats

      目的 探讨大气颗粒物暴露对大鼠心肌组织氧化损伤。
      方法 将48只SPF级SD雄性大鼠随机分为暴露组(3个月,6个月)和对照组,每组12只。对照组大鼠饲养在过滤大气颗粒物的独立通气笼盒(IVC)内,暴露组大鼠饲养在未过滤大气颗粒物的IVC内。HE染色观察6个月后大鼠心肌组织病理学改变。ELISA检测3个月和6个月大鼠心肌组织谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、髓过氧化物歧化酶(MPO)活力和丙二醛(MDA)含量;Western blot法检测大鼠心肌组织白细胞介素-1β(IL-1β)、白细胞介素-8(IL-8)和血管细胞粘附分子-1(VCAM-1)蛋白表达水平。
      结果 与对照组比较,6个月暴露组心肌组织细胞排列紊乱,大量炎性细胞浸润,3个月暴露组GSH-Px活力下降,MDA含量升高,IL-8及VCAM-1蛋白表达升高,6个月暴露组MPO活力和MDA含量上升,IL-1β及IL-8蛋白表达升高,差异均有统计学意义(P < 0.05)。
      结论 大气颗粒物暴露可导致大鼠心肌组织发生氧化损伤,炎症因子蛋白表达水平升高。

       

      Abstract:
      Objective To explore oxidative injury of atmospheric particulate matter exposure on myocardium tissue of rats.
      Methods A total of 48 specific pathogen-free male Sprague-Dawley rats were randomly divided into control group (3 months, n=12; 6 months, n=12) and exposure group (3 months, n=12; 6 months, n=12). Rats in the control group were raised in individually ventilated cages (IVCs) in which atmospheric PM was filtered, while rats in the exposure group were raised in IVCs in which atmospheric PM was not filtered. Hematoxylin-eosin staining was used to observe the pathological changes of rat's myocardial tissue after 6 months. After 3 and 6 months, enzyme-linked immunosorbent assay was used to measure the levels of glutathione peroxidase(GSH-Px), superoxide dismutase (SOD), myeloperoxidase (MPO) and malondialdehyde (MDA) in myocardial tissue of the rats, and Western blot was used to measure the protein expression levels of interleukin-1β (IL-1β), interleukin-8 (IL-8), and vascular cell adhesion molecule-1 (VCAM-1) in the rat's myocardial tissue.
      Results The cells of myocardial tissue in the 6-month exposure group showed a disordered arrangement and massive inflammatory cell infiltration under the light microscope. Compared with the control group, the 3-month exposure group had a significantly decreased activity of GSH-Px and significantly increased MDA level and IL-8 and VCAM-1 protein expression (all P < 0.05); the 6-month exposure group had significantly increased MPO activity, MDA level, and IL-8 and IL-1β protein expression (P < 0.05).
      Conclusion Atmospheric PM exposure can lead to oxidative stress injury in myocardial tissue of the rats and an increase in the expression of inflammatory factors.
