    反相HPLC测定盐酸氨基葡萄糖含量

    Determination of Glucosamine Hydrochloride by HPLC

    • 摘要:
      目的  建立反相HPLC法测定盐酸氨基葡萄糖, 避免目标峰周围出现负峰、与杂质峰分离不好和目标物不保留的问题。
      方法  采用反相高效液相色谱法, 样品经流动相超声提取, 用hilic色谱柱Zorbax Rx-Sil (2.1 mm×150 mm, 5 μm) 分离; 色谱条件为流动相:10 mmoL乙酸铵乙腈 (Ⅴ/Ⅴ=12/88);流速:0.7 mL/min; 检测波长:195 nm; 柱温:30℃。
      结果  本品在0.2~1.0 mg/mL范围内呈线性关系, 相关系数为0.9994。平均回收率为94.2%~96.3%, 精密度为1.21%~2.10%。
      结论  此方法可定量测定胶囊中盐酸氨基葡萄糖, 改进国标方法中目标物不保留的现象, 方法简单, 结果可靠。

       

      Abstract:
      Objectives  To establish a RP-HPLC method for the determination of glucosamine hydrochloride to avoid some problems which might cause negative peaks around the peak, bad separation and object peak not retained.
      Methods  Samples were extracted by ultrasonic wave; the spectrophotometric column was hilic Zorbax Rx-Sil (2.1 mm×150 mm, 5 μm); the mobile phase was 10 mmol/L ammonium acetate-acetonitrile (12:88); the flow rate was 0.7 mL/ min; the detection wavelength was 195 nm; and the column temperature was 30℃.
      Results  The linear range of glucosamine hydrochloride in this product was 0.2~1.0 mg/mL and the correlation coefficient was 0.9994. The average recovery was 93.8%~96.7%, RSD was 1.02%~2.03%.
      Conclusion  The content of glucosamine hydrochloride in glucosamine hydrochloride capsule can be determined simply and accurately by this method.
